Receiving and releasing a culture into routine work

  1. Verify labels, shipping condition, documentation and the expected storage state before opening or thawing.
  2. Create a traceable record linking the vial, source document, operator, date and intended quarantine location.
  3. Recover the culture using the exact source instructions and avoid introducing unplanned optimization variables during the first recovery.
  4. Keep the culture separated until required identity and contamination checks are complete.
  5. Release it into routine use only when the predefined quality criteria are met.

Routine observation is part of the protocol

A media change or passage should not be performed only because a calendar reminder appeared. The operator should inspect the culture and record the evidence that supports the action.

Useful observations include distribution across the vessel, attachment, morphology, medium appearance, debris, growth since the previous observation and any difference between vessels prepared together.

  • Compare cultures at matched time points after seeding.
  • Record deviations before attempting to correct them.
  • Do not describe a culture as “normal” without linking that judgment to its own documented baseline.

Passaging should preserve comparability

The objective is not merely to detach cells and move them. It is to produce a new culture whose starting condition is known and whose recovery can be compared with prior passages.

Document the vessel, observed state before passage, dissociation method, exposure or handling deviations, cell count and viability method, seeding calculation, final volume and time returned to controlled conditions.

Build a working bank before the culture becomes irreplaceable

When identity and contamination status are acceptable, preserve a documented bank according to the cell source and institutional procedure. A working bank reduces repeated genetic and phenotypic drift from indefinite serial passage and provides a known recovery point after an incident.

The freeze record should connect each vial to the parent culture, passage information, freezing medium, controlled cooling method, storage location and release test results.

Boundary note: Cryopreservation formulations and cooling conditions can be cell-type specific. Follow the exact validated procedure rather than copying a generic recipe.
